There is a Total Wine in Laurel, MD not too far from I-95 as the crow flies, although it will take you 25-30 minutes in the car to drive there from the exit. Heavy Seas Brewery is somewhere south of Bawlmer but I'm not sure how close they are to I-95. State Line is the one that draws in the beer geeks, though, and its only 2 minutes off the interstate.
3 miles west of intersection with 84 in New York, just off the Brewster exit of 84, is DeCicco's, a world class bottle store/gourmet supermarket that inside has a world class pub that also counter pressure fills growlers to go of all its selections.
This, and they have pisser, and it's easy to avoid the $6 toll, sign up in advance for their email newletter and get discounts in the store. Somewhat new is also growler fills. Total Wine Alexandria, VA about 1-2 minutes of 395
In Philly Yards and Philadelphia Brewing Co are right off of 95. River Horse is off of 95 in Ewing, NJ
I'll second State Line Liquors in MD. The next border up, in Delaware right before the PA border, is Total Wine in Claymont DE. http://www.beeradvocate.com/beer/profile/4273/?view=ratings Delaware has lower liquor taxes, and no sales tax, so prices are good. It's 30 seconds off the Naamans Road exit of I-95. Going North, this requires you to stay on I-95 through Wilmington, do not take the I-495 bypass or there is no direct Naamans Road exit. Going South, the exit is right at the split.

Great store, but way off 95. The Ardsley location would be closer, but it's still about 30 minutes. To OP, not the mid-Atlantic, but in Branford, CT there is a store off exit 54 called Coastal Wine and Spirits. Pretty large selection of whatever is usually available in Connecticut.
I just stopped at The Perfect Pour south of Baltimore and it was awesome, I'll definitely be going there any time I'm in the area. Its right off 95 too.
Came here to suggest this one. Also in that area is Frisco Taphouse, Pub Dog, and Columbia Alehouse. Frisco is Push Brewing and Columbia Alehouse is Oliver's. Both are pretty good brewpubs. Pub Dog serves their own stuff too, and is a good pizza place. Edit: Also wanna add that Heavy Seas brewery is a few minutes off I95. So that's a quick and easy stop as well.
State Line Liquors is the only store you need to stop at. Easy off, easy on, awesome selection, good prices. I will probably never drive by Elkton again without stopping here. Worth it every single time.
Port Chester Beer Distributor in Port Chester, ny is another good option. They are close to 95, near the ny/ct border. I haven't been there in a while but they had growler fills about 11 months ago.
Stateline is THE place off 95 heading north (as others have mentioned) but if you are in NJ, you may want to check out Joe Canals in Woodbridge.
